Information about Dydrogesterone (Generic Duphaston) :
Duphaston is an active progestogen to be taken orally. Its action takes place directly on the uterus, producing a complete secretory endometrium in an estrogen-primed uterus. You should know that at therapeutic levels, Duphaston has no contraceptive effect. It does not influence ovulation or the corpus luteum. This medicine is non-androgenic, non-estrogenic, non-corticoid, non-anabolic. Duphaston is not excreted as pregnanediol.
Duphaston in combination with an estrogenic substance may be administered to treat secondary amenorrhoea, dysfunctional uterine bleeding and post-menopausal complaints where endogenous progesterone deficiency is implicated.
In case you have irregular cycles due to endogenous progesterone deficiency, especially in irregular cycles due to shortened luteal phase (ie pre-menopause), take Duphaston 5 to 10 mg. You may need several cycles of treatment.
In secondary amenorrhoea
It is recommended to use Duphaston in combination with an estrogen, as in these conditions endogenous progesterone deficiency is usually accompanied by estrogen deficiency. You should take 0,05 mg ethinylestradiol each day from the 1st to the 25th day of the cycle, and 5 mg Duphaston is added twice daily from the 11th to the 25th day. Five days after the subsequent withdrawal bleeding, the same is repeated to imitate a natural cycle.
In dysfunctional uterine bleeding
For the purpose of stopping bleeding: Duphaston 10 mg in combination with 0,10 mg ethinylestradiol twice daily for 5 to 7 days.
For the purpose of preventing heavy bleedings: Duphaston 5 mg twice daily from day 11 to day 25 of the cycle, if necessary, in combination with an estrogen during the first half of the cycle.
In post-menopausal complaints
In case for the purpose of symptomatic treatment of post-menopausal complaints estrogens are already used (hormone replacement therapy HRT), Duphaston 10 mg is taken to counteract the effects of unopposed estrogens on the endometrium. A subsequent withdrawal bleeding is induced.
In case on continuous estrogen therapy: Duphaston 10 mg twice a day during the first 12 to 14 days of each calendar month.
In case on cyclic estrogen therapy: Duphaston 10 mg twice a day during the last 12 to 14 days of the treatment.
Do not use Duphaston in patients with undiagnosed vaginal bleeding or patients with a history of thromboembolic disorders.
Use Duphaston with extreme caution in patients suffer from cardiovascular, renal or hepatic impairment, diabetes mellitus, asthma, epilepsy and migraine.
Use Duphaston with care in patients with a history of mental depression.
Duphaston should be kept in a dry, dark place at temperatures not exceeding 25°C. It should be kept out of reach of children.
Possible side effects
You should consult with your doctor in case you have any of side effects especially bothersome listed below: gastro-intestinal disturbances, allergic skin rashes or urticaria, changes in libido, acne, fluid retention, mass gain, mental depression and breast changes, which may include discomfort or gynaecomastia, virilising, breakthrough bleeding, jaundice.
Be aware that Duphaston may interact with some medicines. Consult with your health care provider in case you are taking any other drugs.
